Emergency services were called at just after 8am following the collision between a VW Golf and a Volvo HGV on Hall Lane at Wrightington near to the hospital.
The car, which was travelling East, appears to have failed to negotiate a bend and crossed into the path of the HGV in the opposite carriageway where they collided head on.
The driver of the Golf, a man in his 70s, and the passenger, a woman in her 60s, both suffered serious injuries and both died.
The driver of the HGV was left shocked but uninjured. He is being treated as a witness to the incident.
The road was closed while police dealt with the incident but is now back open.
Senior Investigating Officer, Sgt Martin Wilcock, said: “Very sadly this collision has resulted in two people losing their lives and my thoughts are with their loved ones. I would appeal to anyone who saw what happened and who has not yet spoken with police, or anyone with footage such as dashcam, CCTV or Ring doorbell to get in touch.
“I would also like to speak to anyone who saw the VW Golf driving in the moments before the collision.
